Des scientifiques am\u00e9ricains ont constat\u00e9 que l\u2019\u00e9limination des chromosomes suppl\u00e9mentaires des cellules canc\u00e9reuses, \u00e0 l\u2019aide de la technique d\u2019\u00e9dition g\u00e9n\u00e9tique CRISPR, permettait d\u2019emp\u00eacher leur d\u00e9veloppement incontr\u00f4l\u00e9.<\/p>\n<p>DES CHROMOSOMES SUPPL\u00c9MENTAIRES<br \/>\nSi les cellules humaines saines poss\u00e8dent 23 paires de chromosomes, les observations r\u00e9alis\u00e9es au cours des derni\u00e8res d\u00e9cennies ont montr\u00e9 que la majorit\u00e9 de leurs homologues canc\u00e9reuses en renferment davantage. Cette anomalie est connue sous le nom d\u2019aneuplo\u00efdie, mais son r\u00f4le exact dans le cancer restait jusqu\u2019\u00e0 pr\u00e9sent obscur : s\u2019agissait-il d\u2019une cause fondamentale ou d\u2019une cons\u00e9quence de la maladie ?<\/p>\n<p>Dans le cadre de travaux publi\u00e9s dans la revue Science, Jason Sheltzer et ses coll\u00e8gues de l\u2019universit\u00e9 Yale se sont concentr\u00e9s sur un type d\u2019aneuplo\u00efdie dans lequel une cellule acquiert une troisi\u00e8me copie d\u2019une structure appel\u00e9e \u00ab bras long \u00bb sur le chromosome 1. Un ph\u00e9nom\u00e8ne observ\u00e9 dans de nombreux types de cancer \u00e0 un stade pr\u00e9coce et li\u00e9 \u00e0 la progression de la maladie.<\/p>\n<p>Lorsque l\u2019\u00e9quipe a utilis\u00e9 son outil d\u2019\u00e9dition g\u00e9n\u00e9tique ReDACT (Restoring Disomy in Aneuploid cells using CRISPR Targeting) pour \u00e9liminer ces chromosomes suppl\u00e9mentaires, elle a constat\u00e9 que les cellules canc\u00e9reuses perdaient la capacit\u00e9 de former des tumeurs malignes.
