Une \u00e9ni\u00e8me tentative qui vise \u00e0 ressusciter cette esp\u00e8ce embl\u00e9matique.<\/p>\n<p>Le retour du mammouth est un mythe toujours bien vivace et les scientifiques n&rsquo;en finissent plus d&rsquo;explorer de nouvelles pistes pour y parvenir. La derni\u00e8re annonce en date vient de chercheurs de l&rsquo;universit\u00e9 de Kindai, au Japon, qui affirment avoir raviv\u00e9 une activit\u00e9 biologique dans des anciennes cellules de mammouths.<\/p>\n<p>Un tout d\u00e9but de division cellulaire<br \/>\nYamagata et son \u00e9quipe ont travaill\u00e9 sur des cellules issues de Yuka, un mammouth laineux mort il y a 28.000 ans et d\u00e9couvert en 2010 dans le perg\u00e9lisol de la Sib\u00e9rie. Ils ont extrait, du tissu musculaire de l&rsquo;animal, 88 noyaux de cellules dont la structure n&rsquo;\u00e9tait pas trop endommag\u00e9e et les ont implant\u00e9s dans des ovocytes de souris. Banco : cinq des cellules incub\u00e9es ont pr\u00e9sent\u00e9 \u00ab des r\u00e9actions biologiques caract\u00e9ristiques du d\u00e9but de la division cellulaire \u00bb, selon Kei Miyamoto, un des membres de l&rsquo;\u00e9quipe. Plus pr\u00e9cis\u00e9ment, les scientifiques ont pu observer un alignement des filaments d&rsquo;ADN, l&rsquo;incorporation d&rsquo;histones (des prot\u00e9ines autour desquelles s&rsquo;enroule l&rsquo;ADN) et une formation nucl\u00e9aire partielle.<\/p>\n<p>Une approche qui avait \u00e9chou\u00e9 il y a 10 ans<br \/>\nOn est bien loin d&rsquo;avoir ressuscit\u00e9 des cellules de mammouth capables de se diviser et de se reproduire, reconnaissent eux-m\u00eames les chercheurs, dont les r\u00e9sultats ont \u00e9t\u00e9 publi\u00e9s dans la revue Scientific Reports le 11 mars dernier. \u00ab Mais cela montre que malgr\u00e9 les ann\u00e9es, une activit\u00e9 cellulaire peut encore se produire \u00bb, se r\u00e9jouit Kei Miyamoto dans un entretien \u00e0 l&rsquo;AFP, qui esp\u00e8re atteindre le stade de la division cellulaire si des noyaux en meilleur \u00e9tat sont trouv\u00e9s. Soutenu par l&rsquo;organisation Revive &amp; Restore, qui vise \u00e0 restaurer des esp\u00e8ces \u00e9teintes, le professeur George Church assure avoir d\u00e9j\u00e0 extrait 44 g\u00e8nes de mammouth, gr\u00e2ce \u00e0 CRISPR\/Cas9, pour pouvoir remplacer certaines parties du g\u00e9nome de l&rsquo;\u00e9l\u00e9phant.<\/p>\n<p>En Russie, une autre \u00e9quipe men\u00e9e par Semyon Grigoriev et Hwang Woo-Suk (pionnier sud-cor\u00e9en du clonage et inculp\u00e9 en 2006 pour fraude, d\u00e9tournement de fonds et violation de lois sur la bio\u00e9thique), tente de recoller des morceaux d&rsquo;ADN de mammouth pour recr\u00e9er un g\u00e9nome artificiel.
